Disintegration and Dissolution in Pressed Candy OEM: Testing, Limits and Formula Fixes

Whether a functional tablet disintegrates in time and releases its actives as intended decides whether it merely looks compliant or actually performs. Disintegration and dissolution answer two different questions: the first asks how quickly a tablet breaks into granules under defined conditions, the second how quickly the active enters solution. For a functional tablet, failing disintegration almost guarantees failing dissolution, but passing disintegration does not guarantee good dissolution - core structure, inter-granule bonding and any coating all rewrite the release curve. In pressed candy contract manufacturing, both parameters belong in formulation design, not in a rework meeting after finished-goods testing fails. This article covers how the two relate, the structural factors behind them, dissolution method design, formulation and process fixes, data interpretation, and common misconceptions.

Disintegration and Dissolution in Pressed Candy OEM: Testing, Limits and Formula Fixes

How disintegration and dissolution relate

Disintegration is the precondition for dissolution. The tablet takes up water, swells, the bonds between granules fail and it breaks into fine particles; those particles then de-aggregate and wet out, and only then does the active enter solution. In theory, the more complete the breakdown, the greater the exposed surface area and the faster the release. The two are not always in step, however. A formula can disintegrate in two or three minutes yet release slowly because the active is hydrophobic, encapsulated by excipients, or trapped behind a high-viscosity gel layer. Equally, a tablet that disintegrates a little more slowly but disperses well can give a smoother curve. Read the two data sets together: passing disintegration is necessary, not sufficient. Set limits to match the product's positioning - food-type tablets are judged on disintegration and mouthfeel, while functional tablets need a dissolution profile under control, with fast or modified release stated explicitly.

Three structural layers that set disintegration

Disintegration speed is decided at three levels. The first is inside the granule: the bond strength between the main ingredients and the binder. In wet granulation, high binder levels and high drying temperatures both harden the granule and slow breakdown. The second is between granules: compression force and lubricant level. Higher compression means lower porosity and slower water ingress, while excess hydrophobic lubricant forms a water-repellent film over the granule surface. The third sits outside the core, where a coating and a polishing layer add a further barrier; a gastric-soluble coat has limited effect, whereas modified-release or enteric coats are deliberate design. Troubleshoot from the outside in: confirm coating and polish first, then force and lubricant, and only then go back to granulation. Of the three, compression force is the easiest to drift, because it can be adjusted on the line at any time.

Designing the dissolution method

Dissolution conditions must be written into the method, or the data cannot be compared across batches. First, the medium: choose water, simulated gastric fluid or simulated intestinal fluid according to positioning, and state whether enzymes are added and whether degassing is required. Second, volume and temperature - 900 mL at 37 C is typical; too small saturates early, too large may fall below the detection limit. Third, the apparatus and speed: an excessive paddle speed can mask a weak formula, while a very low speed can manufacture a failure. Fourth, sampling points - immediate-release products usually take 15, 30 and 45 minutes, modified-release needs fuller coverage. Fifth, the acceptance approach: beyond single-point limits, compare profiles across batches or before and after a change using a similarity factor. Once fixed, the method should not be adjusted to accommodate a batch that fails.

Formulation and process fixes

Adjustments only make sense once you know whether disintegration or dissolution is the failing step. If disintegration is slow, start with the disintegrant: choose one with strong wicking and swelling, and consider adding part of it extra-granularly, which acts faster than a wholly intra-granular addition. Reduce binder level, relax drying temperature, and bring compression force back into the validated window. If disintegration passes but dissolution falls short, the usual cause is poor wetting of the active; a surfactant can improve wetting, while micronisation or pre-blending with a hydrophilic carrier raises dispersibility. If the curve starts fast and flattens, suspect a gel barrier from a high-viscosity excipient and switch to a lower-viscosity grade or reduce its level. Change one variable at a time and re-test disintegration, hardness and dissolution together, otherwise the result cannot be attributed.

Testing and data interpretation

Both tests need disciplined equipment and operation. On the disintegration tester, check basket rise-and-fall frequency, medium temperature and whether discs are used, and keep the number and position of tablets fixed. The dissolution tester needs periodic mechanical verification covering speed, shaft or basket height and centring, and medium volume should be measured rather than read off a mark. Start interpretation with individual variation: if one or two of six tablets lag clearly, weight or hardness variation is the likely cause, and the fix belongs in compression rather than the formula. Then look at batch-to-batch consistency - three consecutive batches on the same formula should overlap broadly before the process is called stable. Retain raw data, curves and instrument parameters; they serve internal traceability and are among the first things a customer audit asks for.

Four misconceptions to avoid

The first is treating disintegration time as the only indicator: a tablet can disintegrate well while the active stays locked inside granules, and the reverse also happens. The second is chasing the formula while ignoring compression force and machine condition - the same formula behaves differently on different presses and at different forces, so confirm equipment and settings first. The third is loosening the dissolution method to accommodate a failing batch; once sampling times or speed are relaxed, historical data loses its comparability. The fourth is reading in-vitro results as in-vivo outcomes: dissolution is a quality-control tool, not an efficacy claim, and the label must stay inside that boundary.

Disintegration and dissolution are two faces of the same design. Fix the disintegrant system, bond strength and coating choice at formulation stage, hold force and lubricant inside the validated window on the line, and accumulate data with fixed methods and fixed sampling points; the product then has both a stable release pattern and a traceable quality record. For a contract manufacturing project those records are also the strongest material you have in customer discussions and audits.
