Transport and Shelf Life in Oral Liquid OEM: Freeze-Thaw Testing

Laboratory stability studies run at constant temperature and humidity, and even accelerated studies merely raise the temperature by a set number of degrees. The journey a product actually makes is far more varied. A case of oral liquid may sit overnight in a cold northern truck, then cross a tropical shipping lane in a container, gaining and losing heat several times along the way. None of this appears in a routine stability report, yet it decides whether the product is still usable on arrival. In oral liquid OEM, transport validation is the step export and multi-region sales must add.

Transport and Shelf Life in Oral Liquid OEM: Freeze-Thaw Testing

Why ambient data is not enough

A routine stability study answers how long a product changes under constant conditions. Transport validation answers whether it changes under varying ones. The difference is that the latter involves compound variables: fluctuating temperature, indefinite duration, and often vibration and compression alongside. More importantly, some changes happen only when temperature crosses a threshold. Certain components crystallise out at low temperature and may not fully redissolve on warming; some hydrocolloid networks are disrupted by freezing and never recover their original viscosity or suspending power. These are irreversible, which constant-temperature studies rarely expose, and once they occur in distribution the product cannot be brought back by simply warming it up.

What freezing does

At low temperature water freezes and expands, loading the container outwards, with the bottle and closure taking the strain first. During freezing, solutes are pushed into the unfrozen region and concentrate locally; unless mixing is thorough after thawing, this leaves uneven strength and taste. In formulas built on hydrocolloids and emulsions, ice crystal growth damages the network and interfacial films, appearing after thaw as syneresis, separation or reduced viscosity. With active ingredients, repeated freeze-thaw can also accelerate degradation or aggregation. How far these changes go depends on freezing rate, minimum temperature, number of cycles and the formula itself, so validation has to use real cycling rather than inference from a single freeze.

The effect of hot routes

Heat causes a different set of problems. Higher temperature accelerates chemical reactions including oxidation, hydrolysis and browning, and speeds the loss of volatile flavour compounds and the degradation of certain actives. Inside a container in direct sun, temperature can run well above ambient, and the day-night swing makes internal pressure rise and fall repeatedly, fatiguing closures and gaskets. For protein or hydrocolloid systems, sustained heat can cause irreversible aggregation or gelation. Heat effects are usually coupled with time, so a short hot spell may be acceptable while a longer one changes the product sharply; assessment therefore needs both peak temperature and duration.

How to run a freeze-thaw study

The basic method holds the product at low temperature for a set period, moves it to ambient or warm conditions to thaw, and repeats the cycle a defined number of times. After each cycle, inspect appearance, measure viscosity and pH, check for sediment and separation, and assay key components. The number of cycles should reflect the real route: three to five may cover multi-region distribution, while exports to cold regions need more. Containers, fill volume and closure must match production or the results mean nothing. The key question is not just whether something changed but whether the change is reversible. Returning to normal after thaw is acceptable; persistent sediment or separation means the formula or pack needs work.

Packaging as part of the answer

Packaging carries a good share of the protection in transit. Bottle material must be chosen for low-temperature brittleness and dimensional stability when warm; some materials crack or deform under freezing expansion. Closures and gaskets must hold their seal through thermal cycling rather than weeping as pressure changes. Outer packaging with cushioning reduces the effect of vibration and stacking load on the bottles. For long routes, a temperature indicator label can record whether the agreed range was exceeded, which is valuable evidence if a claim arises. Once the pack is fixed, any change should prompt a fresh transport assessment, not just an appearance check.

Pass criteria and shipping conditions

Transport validation needs explicit pass criteria, or the study finishes without a release decision. Criteria usually sit at three levels. Sensory: appearance, odour, taste and structure show no significant change. Physical and chemical: viscosity, pH, soluble solids and key actives stay within permitted variation. Packaging: no leakage, deformation or seal failure. All three must pass before the product is considered stable on that route. The results should also feed back into shipping conditions as a written temperature range and time limit, stated in the contract or shipping documents, so a damage claim has an agreed basis for judgement.

Transport stability extends the laboratory concept of stability into real distribution. Ambient data says how long a product lasts under ideal conditions; freeze-thaw and heat studies say what it can survive in practice. Together they support a reliable view of shelf life and sales radius. For export or cross-climate projects this validation belongs in the plan from the moment the product is finalised, rather than being added after complaints arrive.
